Applications of BBa_K5103000

Everything went as expected in regard with using this fragment, digesting, and cloning. We successfully cloned Cry8Da into PCG004 using BsaI cut sites. We also successfully combined Cry8Da and mScarlet fluorescent tag using the HindIII and XhoI cut site towards the 3’ end of Cry8Da right upstream of BsaI. This allowed us to flank the Cry-mScar sequence using BsaI. However, inserting that combined fragment into PCG004 failed and we decided to scratch the idea of using mScarlet entirely.

With regards to iGEM Guelph's 2024 project, project Ash Guard, this part was very well thought out and we successfully used it as we intended to in a variety of plasmids and combinations. For more details, explore our other parts.
